Scientist/Senior Scientist, Discovery Biology and Proteomics

LifeMine Therapeutics · Watertown, MA · 3 days ago
On-siteScienceFull-time

Key Responsibilities

  • Propose study designs and conduct experiments to evaluate immunomodulatory response mechanisms in vitro and in vivo.
  • Interpret data in the context of disease biology, signaling pathways, and therapeutic hypotheses; translate findings into actionable insights for cross-functional teams.
  • Design and execute bottom-up proteomics workflows including sample preparation (protein digestion, fractionation, enrichment), LC-MS/MS acquisition, and data analysis using tools such as MaxQuant, DIA-NN, Spectronaut, and Skyline.
  • Perform affinity-based pulldown and co-immunoprecipitation experiments coupled with MS to identify protein-protein interactors and characterize target interactomes.
  • Execute CETSA (Cellular Thermal Shift Assay) and related target engagement assays to assess drug-target interaction in cellular contexts.
  • Apply chemical biology tools including activity-based probes, photoaffinity labeling, and chemically modified ligands to support mechanism-of-action studies.
  • Partner closely with biology and translational teams to design experiments that address key program questions and efficiently contribute to clinical studies.
  • Communicate results clearly through presentations, reports, and scientific discussions with internal and external stakeholders.

Requirements

  • Ph.D. in Biochemistry, Chemical Biology, Proteomics, Analytical Chemistry, or a related field.
  • 4+ years of postdoctoral or industry experience with a strong focus on mass spectrometry-based proteomics.
  • Proven hands-on experience with LC-MS/MS instrument operation (orbitrap, tribrid and timsTOF), method development, and troubleshooting.
  • Demonstrated experience in bottom-up proteomics workflows (global, PTM enrichment) from sample preparation through data analysis.
  • Experience with affinity pulldown/AP-MS or interactome profiling experiments is a must.
  • Familiarity with CETSA or other thermal proteome profiling (TPP) approaches.
  • Proficient using chemical biology tools for target identification and MoA studies.
  • Strong biological understanding with the ability to contextualize proteomic findings within cellular and disease biology.
  • Track record of independent experimental design, execution, and troubleshooting.
  • Excellent communication skills; ability to present complex data to diverse audiences.
