Stem Cell Research Technician - 140192
UC San Diego Health Sciences is a $1+ billion organization encompassing 16 academic departments, 2 professional schools, 2 hospitals, and various programs dedicated to fulfilling its missions in research, education, and patient care. The Department of Cellular and Molecular Medicine (CMM) is one of two basic science units in the School of Medicine, where researchers study fundamental cellular processes using techniques ranging from classical genetics to high-throughput genomics and stem cell biology.
About the role
In the Yeo lab, we seek to understand the mechanisms by which gene expression is controlled at the RNA level to maintain proper cell function during human development and aging. We study how dysregulation of these processes leads to developmental and degenerative disorders. The Stem Cell Research Technician will assist with generating and analyzing stem-cell-based models of neurodevelopmental disorders under supervision.
Responsibilities
- Perform routine maintenance of human embryonic and induced pluripotent stem cells and their differentiated progeny using established protocols.
- Conduct CRISPR genome editing and routine differentiation of stem cells toward neuronal cells using established protocols.
- Use standard molecular biology tools to transfect, lentivirally infect, and electroporate cells; isolate RNA, DNA, and protein; and characterize gene expression via Western blot, rt-qPCR, and RNA-seq library preparation.
- Assist in immunocytochemistry and epifluorescence/confocal microscopy characterization of cells.
- Perform molecular biology and biochemical assays as needed.
- Support general lab organization, including cleaning, stocking supplies, and maintaining accurate data records.
- Follow step-based experimental protocols and comply with UCSD safety and biohazardous material guidelines.
Requirements
- Theoretical and/or practical knowledge in cellular and molecular biology, immunology, stem cell biology, or a related field.
- Experience with molecular biology techniques, including PCR, DNA preparation/extraction, and gel electrophoresis.
- Ability to prepare buffers, solutions, and staining solutions; experience with biochemistry and laboratory techniques.
- Proficiency with personal computers and software applications (e.g., MS Word, Excel).
- Experience operating confocal, light, and fluorescence microscopes.
- Ability to maintain effective working relationships in a multicultural lab environment.
- Previous experience in human pluripotent stem cell culture and differentiation.
- Willingness to work with animals, lentivirus, radioactive material, human tissue, and standard hazardous laboratory compounds.
- Flexibility to work evenings and weekends when necessary.
Preferred Qualifications
- Experience with recombinant protein expression, purification, and basic protein binding assays.
Special Conditions
- Job offer contingent upon satisfactory background check results.
- Life-support certifications (if applicable) must include hands-on practice and in-person skills assessment; online-only certification is not acceptable.
