Overview
The Targeted Protein Degradation (TPD) Proteomics Core within the Fischer Lab at Dana-Farber Cancer Institute serves as a resource for the Cancer Biology Department, aiding investigators in answering key research questions using mass spectrometry-based proteomics. We employ quantitative chemoproteomic and interactomic approaches to identify drug targets and are continuing to expand our repertoire to meet the needs of our core users. Our core is equipped with extensive mass spectrometry equipment including timsTOF HT, timsTOF Ultra2 and Orbitrap Eclipse tribrid mass spectrometers.
Responsibilities
- Responsible for cell culture and performing drug treatments
- Perform proteomics sample preparation including cell lysis, digestion, peptide cleanup (e.g., S-Trap, SP3), and labeling (e.g., TMT)
- Run samples on LC-MS systems (e.g., Orbitrap, timsTOF) following standard protocols.
- Support routine QC checks and instrument maintenance under staff supervision
- Maintain accurate and well-organized lab notebooks and sample metadata
- Collaborate with core users to ensure timely sample processing and communication
- Assist in the implementation of new protocols and improvements to existing workflows
- Help train junior staff and contribute to a clean and safe laboratory environment
- Maintains and orders laboratory supplies
- Communicates with purchasing and may be responsible for negotiating with vendors
Qualifications
- Bachelor's degree in biology, chemistry, or related field.
- 1-4 years of experience in a mass spectrometry or molecular biology lab (academic or industry).
- Experience operating Thermo and/or Bruker mass spectrometers is required
- Experience with mammalian cell culture and proteomics sample preparation is preferred
- Experience in mass spectrometry data analysis and quantitative methods
- Knowledge of statistical and computational methods and R programming is preferred
- Knowledge of proteomics data analysis software, including Proteome Discoverer, DIANN, spectronaut
- Strong attention to detail and ability to follow SOPs.
