The Core Program

The core program is to translate AD genetics and functional genomics into human brain cells both in vitro 2D and 3D models using human induced pluripotent stem cells (hiPSCs) from population and isogenic lines generated by CRISPR/Cas9 genome editing and in vivo hiPSC/rodent chimera systems in comparison with post-mortem brain. In particular, the research will focus on deciphering the molecular mechanism of major AD risk variants as well as protective variants, associated genes and identifying targeted therapeutics. Furthermore, the research focusing on human iPSC-glial transplantation in AD animal models followed by in vivo electrophysiological and behavior readouts will enable potential stem cell therapeutics.

Requirements

  • Experienced in handling stem cells (mouse or human iPSCs, embryonic stem cells) and brain cell differentiation (2D or 3D cell cultures)
  • Experienced in electrophysiology (in vivo ephys, Ca imaging, MEA, path clamping)
  • (Plus) CRISPRa/i screens, genome-editing
  • Clear written and oral communication and detailed documentation skills, with the ability to collaborate effectively across disciplines
